What happens to an image when viewed with a microscope?

- Microscope images appear upside down and backwards - Movement appears to be in opposite direction than the actual movement

Where is hyaline cartilage located?

-Covers articular ends of long bones -Most of fetal skeleton -Most of larynx, trachea, nose

Cleaning materials

1. Cotton swabs, lens paper, or tissue paper for lenses 2. Paper towels for body and stage

Cleaning solutions

1. Use soap solution for cleaning the body and stage of the microscope 2. Use alcohol or lens cleaner for cleaning the lenses

Total magnification of image

10X eyepiece X magnification of objective

Always view your slide first with the ______ objective. First lower the objective as close as possible to the stage. As you look through the microscope, slowly move it up and away from the stage using the course adjustment

4X - Then switch to a higher objective and only use the fine focus knob when viewing your sample

Most eukaryotic cells have a diameter of what?

7-20 microns (μm), while prokaryotic cells are smaller (0.2-5 μm) - Red blood cells have an average diameter of 7.2 μm, and are a useful reference for size approximation

How to carry the microscope?

Always carry the microscope with two hands, one hand on the base, and one hand on the arm

Simple columnar epithelium (respiratory)

Ciliated

Examine the lens; the oil should fill the space between the slide or ________________ and the ____________

Coverslip; Lens

Clean the 100X oil immersion lens with the cleaning solution provided immediately after use. What do you have to be careful of?

DO NOT GET OIL ON ANY OTHER OBJECTIVES. It ruins them. Use a fresh piece of Kleenex for each lens.

Perichondrium

Dense irregular connective tissue membrane covering cartilage

Working distance

Distance between objective lens and specimen

Stratified squamous epithelium (nonkeratinized) location

Esophagus, mouth, and vagina

Where is elastic cartilage located?

External ear, epiglottis

While looking through the eyepiece use the _______ focus knob to move the lens up and down in tiny increments; this action should bring the tissue into focus

Fine

Loose areolar CT location

Found around every blood vessel, helping to keep the vessel in place. The tissue is also found around and between most body organs.

Do not use coarse focus knob on what?

High focus

The _____________ the magnification, the less working distance you have

Higher

Where is fibrocartilage located?

Intervertebral discs, pubic symphysis, discs of knee joint

Simple squamous epithelium location

Kidney glomeruli; air sacs of the lungs; lining of heart, blood vessels and lymphatic vessels; lining of ventral body cavity (serosae)

Simple cuboidal epithelium location

Kidney tubules; ducts and secretory portions of small glands; ovary surface

Always clean the lenses, both objective and eyepiece, before starting to use the microscope. A few minutes cleaning saves a lot of frustration later. Only use what?

Kleenex/lens paper to clean the lens, never use paper towels. Paper towels are abrasive and can scratch the lens. Wipe the dust off the condenser and light source. Repeat the procedure when you are finished. Don't forget to wipe the stage off.

Stratified cuboidal epithelium location

Largest ducts of sweat glands, mammary glands, and salivary glands

When you move the slide to the right, the image goes to the __________

Left

Clean the oil immersion objective with what?

Lens paper and lens cleaner

Stratified transitional epithelium location

Lines urinary bladder

Stratified columnar epithelium location

Rare in the body; small amounts in male urethra and in large ducts of some glands

Pseudostratified epithelium location

Respiratory tract and portions of male urethra

Stratified squamous epithelium (keratinized) location

Skin, hair, and nails

Always use a coverslip when viewing __________________ to prevent fluid from getting in the objectives, and ruining them

Wet mounts
