Describe the general features of: Monocytes.

Are largest circulating leukocytes, 2-3X the size of surrounding RBC's. they are some clear, light blue cytoplasm surrounding a nucleus that's bi-lobed. they transform into macrophages which then enter other body tissues.

How is a Differential White Cell Count performed?

Examine your slide using the high dry objective.. then apply oil and carefully rotate the oil objective lens into place, adjust with fine adjustment knob and iris diaphragm. Perform tally of WBC'S by marking off each one that you see by type. Each tally will represent one percentage point.

Describe the general features of: Eosinophils

Have an affinity for the Eosin (alkaline) in Wright stain. Granules are in pink-red range.

Describe the general features of: Basophils

Have an affinity for the Methylene blue (which is "basic"). Their large, chunky granules are blue-purple in color. Granules are dark and large that sometimes it is difficult to see the obscured nucleus of a basophil.

Describe the Wrights Stain procedure.

Lay down paper towels, then add 1-3 drops of Wright Stain to sample of the slide until the blood is completely covered. Let stain sit for 2-3 Minutes, Then add same number of drops of the buffer. Blow on the side to mix the stain and buffer. Let the slide sit until you see metallic greenish film or until 8 mind (do not go past 8 mins). Hold slide down with clothespin over sink and rinse with water. Place slide between bibulous paper and remove clothespin.

Wash slides and wash both hands, clean finger (pinky finger) with alcohol pad. Obtain a drop of blood and immediately apply it towards the end on one side. Without delay use second slide a spreader, pulling blood across to he end of the slide in one smooth motion. Lay down on paper towel and completely dry. If extra blood prop slide up at an angle by placing one end over the spreader slide.
