What is the first experiment with a loop and needle called

Experiment 1: Plate to Slant Transfer

Dispose of slides, swabs, and pipettes in the ________ container.

Sharps

what is Experiment 1 called

What is in the air you breathe?

Disinfectants can destroy __________ __________ and ______________ but may not destroy endospores.

vegetative cells and viruses

refers to the concept that microorganisms are everywhere. It's not entirely true, but microorganisms (especially prokaryotes) do occupy an amazing range of environments. For example, some Archaea live in ocean floor vents where temperatures can exceed boiling and chemicals, poisonous to most forms of life, bubble from the Earth's crust. Other microbes live in solid rock. Some bacteria have even been revived after being suspended in amber (fossilized tree sap) or ice for tens of millions of years!

"Ubiquity of Microorganisms",

Why Practice Aseptic Technique? Three things

1. Contaminate others with microorganisms that could lead to infection. 2. Contaminate themselves with microorganisms that could lead to infection. 3. Contaminate media and/or the microorganisms being studied in the lab.

How long should you take to cool your loop and needle

15-20 seconds

Microorganisms not known to cause disease in healthy adults. Examples: Bacillus subtilis, Serratia marcescens, Staphylococcus saprophyticus Standard microbiological practices are followed. Personal protective equipment (PPE) is not required. A sink must be available for hand washing.

BSL 1

What type of biosafety level is Auburn's labs considered

BSL 1 and BSL 2

Indigenous microorganisms that can lead to diseases of varying severity in healthy adults. Examples: Staphylococcus aureus, Streptococcus pyogenes, Salmonella BSL-1 practices plus: Laboratory access is restricted. PPE must be worn including lab coats and gloves. Eye protection and face shields are worn when needed. Work that may generate aerosols or splashes are performed in a biological safety cabinet. Contaminated waste is autoclaved. The laboratory has self-closing doors. A sink and eyewash are available.

BSL 2

Indigenous or exotic microorganisms that cause serious or potentially lethal disease through respiratory transmission. Examples: Mycobacterium tuberculosis, Bacillus anthracis, Francisella tularensis BSL-2 practices plus: Medical surveillance of laboratory employees. Immunizations may be required. Locking, double door access separates the lab's work area from other parts of the facility. PPE must be worn including lab coats, gloves, eye protection, and respirators. All work with microorganisms must be performed in a biosafety cabinet. Decontamination of all waste and lab clothing before laundering. Laboratory must have negative air-flow, and exhaust air cannot be recirculated. A hands-free sink and eyewash must be available

BSL 3

Microorganisms that are dangerous and exotic with high risk of aerosol transmitted infections. Rarely are there treatments or vaccines for these microorganisms, and the diseases they cause are frequently fatal. Examples: Ebola virus, Marburg virus BSL-3 practices plus: Change clothing before entering lab. Shower upon exiting lab. Decontaminate all materials before exiting lab. All work with microorganisms must be performed in a Class III biosafety cabinet or by wearing a full body, positive pressure suit. Laboratory must have negative air-flow, and exhaust air cannot be recirculated. The laboratory must be in a separate building or in an isolated and restricted zone of the building

BSL 4

All items that have come in contact with bacteria are disposed in the _______ can (blue bag).

Biohazard

What is the second experiment called with the loop and needle

Experiment 2: Broth to Plate Transfer

what is experiment 2 called

Experiment 2: What is on the surfaces you touch?

What is the third experiment called with the loop and needle

Experiment 3: Plate to Broth Transfer

what is experiment 3 called

Experiment 3: What is on your skin?

what is experiment 4 called

Experiment 4: This is why you don't cough in other people's faces.

True or False You can touch the face, apply cosmetics, adjust contact lenses, or bite nails.

False- Do not touch the face, apply cosmetics, adjust contact lenses, or bite nails

Loops and needles are used to transfer cultures. Prior to use, these utensils must be sterilized. This is accomplished by placing the loop or needle into the Bunsen burner flame until it is red hot. The entire wire portion of the loop or needle must be heated this way

Flaming your loop and needle

Disinfect bench before and after the laboratory session with a disinfectant (________ in this lab).

Lysol

All work surfaces must be wiped down with a disinfectant. _________, 70% __________, 10% _________, and quaternary _________ are all examples of popular disinfectants

Lysol, ethanol, bleach, ammonia

is a general term for living organisms that cannot be seen without a microscope, even though some people include viruses and prions in the group.

Microorganism

What are these listed under? a. Wear closed-toe shoes that cover the top of the foot. b. Wear gloves when handling microorganisms or handling chemicals. Do not wear gloves outside the laboratory. c. Wear lab coat while in the laboratory. Do not wear lab coats outside the laboratory

Personal Protection

Gum, paper products not contaminated with microorganisms, and items brought in to the laboratory are disposed of in the _________ _________ cans.

Regular garbage

True or False- Individuals not enrolled in the course are not allowed in the laboratory.

True

True or false It is absolutely necessary to allow the loop or needle to cool before transferring an inoculum of bacteria

True

True or false you should wash your hands before and entering lab

True

Glassware (flasks, beakers, bottles, test tubes) are placed on the ________ cart.

discard

___________-_____________ students (including those who are pregnant or may become pregnant) and students living with or caring for an ___________-______________ individual should consult a physician to determine the appropriate level of participation in the laboratory.

immune-compromised
